TRH (Protirelin) is a tripeptide hormone synthesized in the hypothalamus that binds to thyrotropin-releasing hormone receptors on the anterior pituitary to stimulate synthesis and release of thyroid-stimulating hormone and prolactin, positioning it as the master regulator of the hypothalamic-pituitary-thyroid axis. With a molecular weight of 362.39 g/mol, it has expanded research roles as a neuromodulator with rapid antidepressant effects in depression models and facilitates motor skill acquisition through cerebellar activity. It reverses respiratory depression in opioid overdose without affecting pain control, improves mean arterial pressure in hypovolemic shock, and mitigates oxidative damage by reducing amyloid plaque buildup and modulating excitatory neurotransmitter release to prevent excitotoxicity.
