PNC-27 is a synthetic peptide containing the HDM2 binding domain and transmembrane domain from the p53 tumor suppressor protein that targets cancer cells overexpressing HDM2 on their cell membrane, binding with high affinity to form stable transmembrane pores leading to rapid membrane lysis and necrotic cell death. With a molecular weight of 4031.7 g/mol, it demonstrates selective toxicity towards a wide range of cancer cell lines including pancreatic, breast, leukemia, melanoma, and ovarian cancer cells while leaving non-transformed cells unharmed. In AML cell lines it induces necrosis without activating apoptotic markers, and the selectivity is driven by the presence of HDM-2 in cancer cell membranes, offering a targeted approach to circumvent systemic toxicity of traditional chemotherapeutics.
