Delta Sleep-Inducing Peptide (DSIP) is a naturally occurring nonapeptide first isolated from rabbit brains during slow-wave sleep, with the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu and a molecular weight of 848.82 g/mol. It influences diverse CNS functions by modulating key neurotransmitter systems, including enhancing GABA’s inhibitory actions, attenuating NMDA excitatory actions, and interacting with opioid receptors. While known for normalizing sleep patterns and improving sleep structure in insomnia models, DSIP also exhibits significant stress-modulatory effects, protects against metabolic stress by promoting oxidative phosphorylation in hypoxic conditions, and has shown potential in addiction research with high recovery rates in ethanol and opiate withdrawal models.
