Syn-Coll (Palmitoyl Tripeptide-5) is a synthetic lipopeptide analog of Thrombospondin-1 that binds to and activates latent TGF-β, stimulating fibroblasts to produce extracellular matrix components, particularly type I and III collagen, while inhibiting MMP-1 and MMP-3 activity responsible for collagen breakdown. With a molecular weight of 611.9 g/mol, it demonstrates 2-3 fold increases in collagen mRNA and is approximately 3.5 times more potent than control peptides in reducing wrinkle depth and skin roughness. It improves skin barrier function by reducing transepidermal water loss, and modified versions with ascorbic acid show additional promise by reducing melanin content in melanocytes while stimulating procollagen synthesis.
