PE-22-28 is a synthetic heptapeptide (Gly-Val-Ser-Trp-Gly-Leu-Arg) and truncated, stable analog of Spadin that acts as a potent and selective antagonist of the TREK-1 potassium channel, highly expressed in brain regions associated with mood regulation. With a molecular weight of 773.89 g/mol, it increases neuronal excitability and promotes neurogenesis, with studies showing it can promote neurogenesis and synaptogenesis in as little as four days through four-fold increases in phosphorylated CREB. It influences serotonin neurotransmission by increasing firing rates of serotonin neurons, may increase muscle contractility through mechano-gated channel blockade, and importantly does not increase seizure susceptibility, potentially increasing resistance to seizures.
