Humanin is a 24-amino-acid peptide uniquely encoded by mitochondrial DNA that functions as a potent cytoprotective protein by directly interfering with the pro-apoptotic protein Bax, preventing mitochondrial disruption and cytochrome c release. With a molecular weight of 2687.3 g/mol, it protects neurons in Alzheimer’s models by binding to formyl peptide receptors to mitigate amyloid-beta toxicity, reduces oxidative stress and restores SOD activity in neurotoxicity models, and decreases myocardial fibrosis by downregulating pro-fibrotic cytokines. It also protects retinal pigment epithelium from oxidative stress, supports bone health by reducing osteoclast formation, and improves insulin sensitivity by activating STAT-3 signaling in the hypothalamus.
